Scene in the Original Work:
In Fullmetal Alchemist, Edward and his brother Alphonse try human transmutation to bring their dead mother back, but it fails. Ed loses his left leg, and Al's whole body is taken. Bleeding, Ed draws a transmutation circle on an old suit of armor and cries that he will give his arms, his legs, even his heart, then shouts this line. He loses his right arm as the price, and Al's soul is bound to the armor.

Cultural Context:
It is the starting point of the Elric brothers' journey, and it is shown again in flashbacks. In the manga it is written "だから!! 返せよ!! たった一人の弟なんだよ!!", and the 2009 anime (Episode 1) uses almost the same words. Fans may also quote it, half-jokingly, when someone takes away something precious, like the last piece of cake.
