In the manga story 「ジャイアンの心の友」 (volume 11), Nobita wants revenge on Gian, but Doraemon suggests being kind to him instead. They bring Gian a gadget that turns his own singing into records. After listening to his songs for ages and even making copies and jackets, they hand him the finished records. Gian is moved and declares that Nobita is his 心の友.

Cultural Context:
Gian (Takeshi Goda) is usually the bully who takes Nobita's things, so his sudden switch to 心の友 is part of the joke. This story is said to be the phrase's first appearance, and afterwards Gian shouts 「心の友よ！」 whenever someone praises his singing or supports his dream of becoming a singer. In 2012, a song titled 「フレンド・オブ・ザ・ハ～ト」, an English version of the phrase, was performed in the anime. It may also be used as a joke to thank a friend who helped you out.

This よ is the old-style calling よ, as in 友よ or 神よ, not the sentence-ending よ that adds emphasis (行くよ). In daily life 心の友 is mostly a joke; for a serious "best friend", 親友 is the usual word.
